A single bio-energetics growth and reproduction model for the oyster Crassostrea gigas in six Atlantic ecosystems ArchiMer
Alunno-bruscia, Marianne; Bourles, Yves; Maurer, Daniele; Robert, Stephane; Mazurie, Joseph; Gangnery, Aline; Goulletquer, Philippe; Pouvreau, Stephane.
Many studies based on bioenergetics growth models have investigated the effects of environmental factors on oyster (Crassostrea gigas) growth and physiology. However, most of these models are site-specific and cannot be applied to other culture sites without the re-estimation of parameters or re-formulation of some processes. We aimed to develop a generic growth model suitable for application in contrasting environments, with a constant set of parameters. We tested the oyster-DEB model (Bourlès et al. 2009) for the stimulation of C. gigas growth in different cohorts (spats and adults) at major shellfish culture sites in France, in several years: Arcachon (1993–1994); Marennes-Oléron (2007); Quiberon (1999, 2000, 2001); Rade de Brest (2008); Baie du...

Trente...

Molecular cloning and seasonal expression of oyster glycogen phosphorylase and glycogen synthase genes ArchiMer
Bacca, Helene; Huvet, Arnaud; Fabioux, Caroline; Daniel, Jean-yves; Delaporte, Maryse; Pouvreau, Stephane; Van Wormhoudt, A; Moal, Jeanne.
To investigate the control at the mRNA level of glycogen metabolism in the cupped oyster Crassostrea gigas, we report in the present paper the cloning and characterization of glycogen phosphorylase and synthase cDNAs (Cg-GPH and Cg-GYS, respectively, transcripts of main enzymes for glycogen use and storage), and their first expression profiles depending on oyster tissues and seasons. A strong expression of both genes was observed in the labial palps and the gonad in accordance with specific cells located in both tissues and ability to store glucose. Cg-GPH expression was also found mainly in muscle suggesting ability to use glycogen as readily available glucose to. supply its activity. For seasonal examinations, expression of Cg-GYS and Cg-GPH genes...

Growth of the Pacific oyster (Crassostrea gigas) in a high-turbidity environment: Comparison of model simulations based on scope for growth and dynamic energy budgets ArchiMer
Barille, Laurent; Lerouxel, Astrid; Dutertre, Mickael; Haure, Joel; Barille, Anne-laure; Pouvreau, Stephane; Alunno-bruscia, Marianne.
We compared growth simulations by dynamic energy budget (DEB) and scope for growth (SFG) models of the Pacific oyster Crassostrea gigas, cultivated in Bourgneuf Bay on the French Atlantic coast. This bay is located at a latitude in the middle of the European range of the species, and is characterized by high concentrations of suspended particulate matter (SPM) and a marked gradient between high-turbidity sites in the north (daily SPM > 500 mg L-1) and intermediate-turbidity sites in the south. The models use two forcing variables: seawater temperature and food density. We tested two indices of food availability: chlorophyll a and microalgal concentrations. In the SFG model, food intake is simulated by a type-II Holling functional response, as in the DEB...

In situ spawning in a marine broadcast spawner, the Pacific oyster Crassostrea gigas : Timing and environmental triggers ArchiMer
Bernard, I.; Massabuau, J. -c.; Ciret, P.; Sow, M.; Sottolichio, Aldo; Pouvreau, Stephane; Tran, D..
The precise environmental conditions under which broadcast spawners spawn in the field remain largely unknown. We investigated this issue in the oyster Crassostrea gigas using three different methods at different time scales in two traditional oyster farming areas of the French Atlantic Coast, the Bay of Arcachon and Marennes-Oléron. We directly recorded spawning at high temporal resolution using high-frequency non-invasive (HFNI) valvometry from 2007–2014 and measured the dry mass and oyster larvae abundance in 2008 and 2009. We analyzed a 29-yr series of oyster D-larvae numbers in the Bay of Arcachon (1982–2010). By combining these three approaches, we demonstrated that during the summer months at both sites, spawning in C. gigas occurs in the morning or...

Bernard, Ismael; Allain, Gwenhael; Auby, Isabelle; Plus, Martin; Drean, Tugdual; Gasmi, Sonia; David, Valérie; Pouvreau, Stephane; Maurer, Daniele.

Effect of phytoplankton and temperature on the reproduction of the Pacific oyster Crassostrea gigas: investigation through DEB theory ArchiMer
Bernard, Ismael; De Kermoysan, Goulwen; Pouvreau, Stephane.
DEB theory can be used to obtain a detailed description of energy allocation in organisms and the control of this allocation by temperature and food concentration. In this study, we modified the model of Bourlès et al. (2009) developed for the Pacific oyster, Crassostrea gigas, to improve the description of reproductive effort. The model was amended in two ways: a new set of parameters was incorporated and a full description of gonad construction in spring was added, with a new state variable. The application of this model to a dataset for oyster growth measured in four bays over two years showed that this model better described reproductive effort, without modifying overall growth dynamics. We then explored the reproductive responses of oysters to their...

Ovarian and sperm regulatory peptides regulate ovulation in the oyster Crassostrea gigas ArchiMer
Bernay, Benoît; Baudy-floc'H, Michele; Zanuttini, Bruno; Zatylny, Céline; Pouvreau, Stephane; Henry, Joël.
For more than six decades, several studies have shown that genital products to entering the mantle cavity via the incurrent siphon, initiate in oyster, strong and rhythmic contractions of the adductor muscle (AM). In order to characterize the regulatory peptides capable of triggering AM contractions, we focused on the identification of putative myotropic peptides from genital products. Two experimental approaches were developed. The first one, based on a mass spectrometry screening of the male genital products, led to the identification of the tetrapeptide APGWamide. This neuropeptide was also detected in the seminal secretions of the cephalopod Sepia officinalis. In this species, APGWamide is directly involved in the oocyte transport. In Crassostrea, in...

Genetic variation and trade-offs for reproduction ans survival in the Pacific oyster Crassostrea gigas ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Pouvreau, Stephane; Normand, Julien; Ernande, Bruno.
To date, the most significant method to genetically reduce reproductive effort in the Pacific oyster (Crassostrea gigas) has been through the production of triploids, especially since the development of tetraploid lines allowing the breeding of 'natural' triploids. Gametogenesis of triploid oysters is strongly reduced compared with diploids, although they are not fully sterile and can produce viable gametes and some progenies when crossed with diploids. Reduced reproductive allocation and higher heterozygocity are commonly proposed as the main reasons why triploids often present superior yield compared with diploids. Temperature and food availability are known to favour gametogenesis in triploids, but the existence of genetic variation for this trait...

Modelling growth and reproduction of the Pacific oyster Crassostrea gigas: Advances in the oyster-DEB model through application to a coastal pond ArchiMer
Bourles, Yves; Alunno-bruscia, Marianne; Pouvreau, Stephane; Tollu, Guillaume; Leguay, Didier; Arnaud, Christophe; Goulletquer, Philippe; Kooijman, S.
A bio-energetic model, based on the DEB theory exists for the Pacific oyster Crassostrea gigas. Pouvreau et al. [Pouvreau, S., Bourles, Y., Lefebvre, S., Gangnery, A., Alunno-Bruscia, M., 2006. Application of a dynamic energy budget model to the Pacific oyster, C. gigas, reared under various environmental conditions. J. Sea Res. 56, 156–167.] successfully applied this model to oysters reared in three environments with no tide and low turbidity, using chlorophyll a concentration as food quantifier. However, the robustness of the oyster-DEB model needs to be validated in varying environments where different food quantifiers reflect the food available for oysters, as is the case in estuaries and most coastal ecosystems. We therefore tested the oyster-DEB...

Modelling Crassostrea gigas growth and reproduction in different contrasted ecosystems by using dynamic energy budgets : generic validation of the oyster-DEB model ArchiMer
Bourles, Yves; Maurer, Daniele; Le Moine, Olivier; Geairon, Philippe; Mazurie, Joseph; Gangnery, Aline; Alunno-bruscia, Marianne; Pouvreau, Stephane; Goulletquer, Philippe.
Oyster farming is the main aquaculture activity in France, where Pacific oyster (Crassostrea gigas) is spatially cultured from the English Channel to the Mediterranean coasts. Oyster growth performance monitored along French coasts are widely heterogeneous among culture sites and over years. Many studies have been carried out to understand the effects of environmental factors on oyster growth and physiology, by using bioenergetics growth models. However, most of these studies were site-specific. As an example, the model developed by Barillé et al. (1997), which was extensively specified with more than 50 parameters, was validated in the Marennes-Oléron Bay, but cannot be successfully applied to other culture sites without re-estimating parameters or...

Particulate matter in Takapoto lagoon waters: potential food for cultivated pearl oysters ArchiMer
Buestel, Dominique; Pouvreau, Stephane.
Results of research concerning the food of the pearl oyster Pinctada mai margaritifera are presented, by taking Takapoto atoll (Tuamotu archipelago, French Polynesia) as a study site. Monthly sampling of several parameters representative of bivalve environment and food (water temperature, salinity, mineral and organic matter, proteins, lipids and carbohydrates, chlorophyll a and phaeopigments) confirm and increase knowledge of the Takapoto lagoon hydrobiology. The variability of these parameters was analysed. Takapoto lagoon showed a remarkable spatial stability. Temporal variations showed a low seasonal trend. Local meteorology (wind, precipitation) had an influence on short-term variability (especially for mineral matter). Night and day rhythm also...

Buestel, Dominique; Pouvreau, Stephane; Tiapari, Jerome; Bougrier, Serge; Chabirand, Jean-michel; Geairon, Philippe; Fougerouse, A.
A study of pearl oysters Pinctada margaritifera growth and mortality was done on a monthly basis between June 1990 and September 1991 in Takapoto atoll (Tuamotu archipelago). Meanwhile, sampling parameters representative of bivalves food showed a remarquable spatial stability whithin the lagoon. The amplitude of temporal variability varied in the order of some days. Carrying capacity appears low : energetic content of particulate material in waters is about 2 joules/liter, ten times lower than in temperate areas. On the opposite, seasonal effect is reduced and particulate food is always available. Oysters rearing conditions were appropriate during the studied period with a low mortality rate and a regular growth in size (from 97 mm to 136 mm). Pearls...

COAST-HF (Coastal OceAn observing SysTem – High Frequency) is a French national observation network of the physical and biogeochemical dynamics of the coastal ocean, at high frequency. COAST-HF aims at understanding and analysing changes of contrasted coastal ecosystems at different temporal scales from extreme or intermittent high frequency (hour, day) events to multi-year trends. Since several years (from 2000 for the longest time series in Bay of Brest), the network extends along the English Channel, Atlantic and Mediterranean French coasts through 14 fixed platforms instrumented for the in situ high-frequency (≤ 1h) observations. Several French research institutes (IFREMER, CNRS, Marine Universities) are operating these systems. The organization of...

Cochet, Hélène; Pouvreau, Stephane; Tanguy, Ludovic; Kerisit, Christophe.

Addressing critical limitations of oyster ( Ostrea edulis ) restoration: Identification of nature‐based substrates for hatchery production and recruitment in the field ArchiMer
Colsoul, Bérenger; Pouvreau, Stephane; Di Poi, Carole; Pouil, Simon; Merk, Verena; Peter, Corina; Boersma, Maarten; Pogoda, Bernadette.
The European flat oyster (Ostrea edulis) is an ecosystem engineer that provides important biogenic reef habitat with associated ecosystem functions and services. Most stocks have been commercially exploited and degraded; some are functionally extinct. Ecological restoration now aims to recover these degraded, damaged or destroyed ecosystems. Availability of seed oysters and substrate for successful larval recruitment has been identified as a major limiting factor for restoration projects in Europe. In substrate‐limited areas, restoration approaches have to involve the restoration of suitable substrates. The present study provides an evaluation of such potential substrate types. Various categories were investigated through hatchery and/or field experiments:...
